Structures of gangliosides from bovine adrenal medulla

Abstract: Five gangliosides, accounting for over 95% of the total ganglioside fraction, were isolated from bovine adrenal medulla by preparative thin-layer chromatography and the carbohydrate structures determined by a combination of periodate oxidation and permethylation techniques. Partially methylated alditol acetates were generated from the neutral sugars of the fully methylated glycolipids and identified by gas-liquid chromatography. Substitution on N-acetylgalactosamine was determined by methanolysis of the permet… Show more

“…medulla, which contains predominantly hematosides (Price et al, 1975;Ariga et al, 1980Ariga et al, , 1982Ariga et al, , 1983). The presence of high levels of brain and sympathetic neuron like polysialogangliosides in PC 12 cultures may therefore reflect the neuronal rather than the chromaffin-like properties of these cells.…”

“…Pi glycolipid and the tetraglycosylceramide resulting from the a-galactosidase reaction were permethylated according to the method of Hakomori (1964), as applied in this laboratory (Kundu et al, 1975a), and the products were purified by preparative TLC. Approximately 50 Hg was subjected to hydrolysis and reduction according to the method of Bjorndal et al (1970) as applied in this laboratory (Price et al, 1975). The mixture was passed through a small column of Dowex 50W (H+) to remove hexosamine-derived products which gave poorly defined peaks on GLC; these sugars were analyzed separately (see below).…”

“…Total lipid-bound sialic acid was measured by the method of Yu & Ledeen ( 1970) and the concentrations of individual gangliosides were determined by the method of Suzuki (1964), as modified by MacMillan & Wherrett (1969). The procedures used for analysis of permethylated neutral sugars and hexosamines have been described in detail (Price et al, 1975;Kundu et al, 1975;Naiki et al, 1975).…”
