Structures and Biological Activities of Peptidoglycans of Listeria monocytogenes and Propionibacterium acnes12

Abstract: The cell-wall skeletons of Listeria monocytogenes strain EGD and Propionibacterium acnes strain C7, which have the ability to induce macrophage activation, were analyzed, and the structures of the peptidoglycans were investigated. The analytical data indicate that both peptidoglycans have glucosamine residues with free amino groups, which are responsible for the resistance to lysozyme. Possible structures of these peptidoglycans were deduced from the composition and the results of determination of N- and C-ter… Show more

“…The partial hydrolysate of the peptidoglycans of P. technicum, P. avidum, P. granulosum, and C. parvum contained several peptides and saccharides, namely AlaGlu, Glu-Agpm, A 2pm-Gly, A 2pm-Ala, Ala-Glu-Ajpm, GlcN-Mur-Ala, and GlcN-Mur, which were similar to those of P. acnes (9). In the case of the peptidoglycans of P. jreudenreichii and P. shermanii, the peptides Ala-Glu, Glu-Ajpm, …”

“…The resistance to degradation was attributed to the unique structure of the peptidoglycan (PGN) component, which has been identified as the active principle of cell wall skeleton preparations of P. acnes (40,41). At present it is not known whether the unique structure of P. acnes PGN is responsible for the TLR9-mediated effects.…”

“…It is a polymer of alternating units of the disaccharide N-acetylmuramic acid (MurNAc)-(␤-1,4)-Nacetylglucosamine (GlcNAc), cross-linked by peptidic bridges. Muropeptides, which in L. monocytogenes are L-alanyl-␥-Dglutamyl-meso-diaminopimelyl-D-alanine-D-alanine, are bound to the MurNAc residue and are connected by a direct link between the D-Ala residue of one lateral peptide and the mesodiaminopimelyl residue of the other stem peptide (44,57,89,162). Partial deacetylation of GlcNAc residues by PgdA enzyme is another characteristic of Listeria peptidoglycan (21,89) (see below).…”
