Structure of homoserineO-acetyltransferase fromStaphylococcus aureus: the first Gram-positive ortholog structure

Abstract: Homoserine O-acetyltransferase (HTA) catalyzes the formation of L-O-acetyl-homoserine from L-homoserine through the transfer of an acetyl group from acetyl-CoA. This is the first committed step required for the biosynthesis of methionine in many fungi, Gram-positive bacteria and some Gram-negative bacteria. The structure of HTA from Staphylococcus aureus (SaHTA) has been determined to a resolution of 2.45 Å. The structure belongs to the α/β-hydrolase superfamily, consisting of two distinct domains: a core α/β-… Show more

“…Est22 forms stable dimers in solution (Fig. 4B), consistent with HTAs and HTA-like acetyl esterases, including MekB and CgHle (13,14,21). The topological structure of Est22 is similar to those of HTAs and HTA-like acetyl esterases (Fig.…”

“…The root mean square deviations between Est22 and reported HTA-like esterases are greater than 1.667 Å. Like HTAs and HTA-like acetyl esterases (14,16,21), monomeric Est22 consists of two domains, i.e., an ␣/␤ hydrolase domain (residues Met 25 to Asp 205 and Phe 313 to Asn 393 ) and a helical bundle domain (residues Ala 206 to Leu 312 ) (Fig. 4D).…”
