Structure of DNA-Cationic Liposome Complexes: DNA Intercalation in Multilamellar Membranes in Distinct Interhelical Packing Regimes

Abstract: Cationic liposomes complexed with DNA (CL-DNA) are promising synthetically based nonviral carriers of DNA vectors for gene therapy. The solution structure of CL-DNA complexes was probed on length scales from subnanometer to micrometer by synchrotron x-ray diffraction and optical microscopy. The addition of either linear lambda-phage or plasmid DNA to CLs resulted in an unexpected topological transition from liposomes to optically birefringent liquid-crystalline condensed globules. X-ray diffraction of the glob… Show more

“…The sequence of events outlined above leads to a strong electrostatic attraction. This type of electrostatic interaction has been observed for self-assembled complexes between membranes and a variety of anionic polymers, including DNA [64][65][66], F-actin [67], microtubules [68], and filamentous phages [69].…”

“…33 The population(s) of complexes, which actually transfect cells is still uncertain with several models proposed, such as beads on a string, 34 spaghetti and meatballs 33 and multilamellar sheets. 35 However, there is also convincing evidence to support the existence of macro-aggregates within each of these populations. 33 Several factors favour macro-aggregate formation such as the impurity of the DNA, the composition of the suspending vehicle and the ratio of cationic liposome to DNA.…”

“…We use liposome-mediated gene transfaction 8,9 to either block maternal Hoxa10 expression with a Hoxa10 antisense oligonucleotide or to increase maternal Hoxa10 expression with a plasmid that constitutively expresses Hoxa10. These experiments demonstrate that manipulation of adult Hoxa10 expression significantly affects fertility.…”
