Structure of CARDS toxin, a unique ADP-ribosylating and vacuolating cytotoxin from Mycoplasma pneumoniae

Abstract: Mycoplasma pneumoniae (Mp) infections cause tracheobronchitis and "walking" pneumonia, and are linked to asthma and other reactive airway diseases. As part of the infectious process, the bacterium expresses a 591-aa virulence factor with both mono-ADP ribosyltransferase (mART) and vacuolating activities known as Community-Acquired Respiratory Distress Syndrome Toxin (CARDS TX). CARDS TX binds to human surfactant protein A and annexin A2 on airway epithelial cells and is internalized, leading to a range of path… Show more

“…Recently, CARDS toxin was recognized by Kannan et al., which is involved in the pathophysiology of Mp disease. The toxin binds to human surfactant protein A and annexin A2 on airway epithelial cells and is internalized, leading to a range of pathogenetic events . Other factors are well known for their harmful effects via oxidative stress.…”

“…The N-terminal region or Domain 1, which is homologous to the pertussis toxin S1 subunit (PTX) from Bordetella pertussis, includes the catalytic ADP-ribosylating region (Becker et al, 2015;Kannan et al, 2005;Kannan et al, 2014). The C-terminal region of CARDS toxin (Domains 2 and 3) contains the cell receptor binding and vacuolating activities .…”
