Structural gene for the phosphate-repressible phosphate-binding protein of Escherichia coli has its own promoter: complete nucleotide sequence of the phoS gene

Abstract: The complete nucleotide sequence of the phoS gene, the structural gene for the phosphate-repressible, periplasmic phosphate-binding protein Escherichia coli K-12, was determined. The phosphate-binding protein is synthesized in a precursor form which includes an additional N-terminal segment containing 25 amino acid residues, with the general characteristics of a signal sequence. The amino acid sequence derived from the nucleotide sequence shows the mature protein to be composed of 321 amino acids with a calcul… Show more

“…This somewhat confusing picture required positive resolution by an unambiguous method, and the DNA sequencing of the entire region was undertaken. The sequence of the phoS gene was reported by Surin et al (33) and by Magota et al (19) with full agreement. The present work completes the nucleotide sequence of the region.…”

“…Four open reading frames were detected beyond the end of the previously described phoS gene (19,33) and within the region specified above. The open reading frames (ORFs) are transcribed in the same direction as the phoS gene (19,33), and each is preceded by a putative ribosome-binding site appropriately spaced (32) from a presumed ATG initiating codon. ORF-1 and ORF-2 have overlapping stop-start codons.…”

“…The mature phosphate-binding protein of E. coli, PstS, contains 321 amino acids and also exists as a precursor form with the first 25 amino acids representing a typical signal peptide (Surin et at., 1984). The pstS gene is part of the phosphate regulon, which in E. coii comprises a set of more than 20 unlinked genes whose promoters can be regulated simultaneously by overlapping and separate controls.…”

“…Probing of Synechococcus sp. WH7803 chromosomal DNA with a 5.4 kb /-//ndlll fragment containing the E. coii phoS gene (now designated pstS) from pAN92 (Surin et at., 1984) yielded no detectable hybridization, even at low stringency. This Would agree with observations by Ray et al (1991), who stated that most genes of the E. coli pho regulon do not hybridize to cyanobacterial DNA.…”
