This study aimed to investigate structural features and antihyperlipidemic effects of the stigma maydis polysaccharide, termed SMP‐1. This polysaccharide was composed of D‐mannose, L‐rhamnose, D‐glucose, D‐galactose, L‐arabinose, D‐xylose, and D‐galacturonic acid, with a molar ratio of 1.00:0.21:1.41:1.44:0.70:0.44:0.56. The SMP‐1 was mainly bonded by (1 → 6) and (1 → 3) linkages, with various monosaccharides being evenly distributed in the main and side chains. Moreover, SMP‐1 had neither triple‐helical structure nor molecular aggregation. Importantly, the SMP‐1 could effectively bind the bile acids in vitro and significantly lower the total cholesterol, triglyceride, low‐density lipoprotein cholesterol levels, and moderately increase the high‐density lipoprotein cholesterol level in poloxamer 407‐induced hyperlipidemic mice. Moreover, pretreatment with SMP‐1 (≥300 mg/kg) could remarkably reduce fat accumulation and restore hepatocyte morphology in the liver of hyperlipidemic mice. Altogether, these findings indicated that SMP‐1 could be developed as a safe and effective food supplement for preventing and treating hyperlipidemic disorders.

Summary In this study, we compared the chemical structure, antioxidant and immuno‐stimulating activities of two polysaccharides, TPS–FJ and TPS–SC, extracted from Tremella fuciformis from two of the major cultivation regions of China. TPS–FJ and TPS–SC contained protein and high uronic acid. Average molecular weights of TPS–FJ and TPS–SC were 45 461 and 25 981 kDa, respectively. A triple‐helix conformation was exhibited by TPS–FJ but not by TPS–SC. TPS–FJ and TPS–SC contained same monosaccharides but in different ratios. Both the polysaccharides displayed characteristic polysaccharide bands in their Fourier transform infrared and NMR spectra, shown high water and oil holding capacity, and retained antioxidant potential after simulated gastrointestinal digestion in vitro. TPS–FJ significantly (P < 0.05) stimulated the nitric oxide, tumour necrosis factor‐α and interleukin‐6 production of RAW 264.7 cells with higher reactions than those of TPS–SC in vitro. Thus, Tremella fuciformis polysaccharide can be used as a potential antioxidant and immunomodulatory ingredient in food industry.

Preparation, Characterization, and Immuno-Enhancing Activity of Polysaccharides from Glycyrrhiza uralensis

Glycyrrhiza uralensis is a Chinese herbal medicine with various bioactivities. Three fractions (GUPS-I, GUPS-II and GUPS-III) of G. uralensis polysaccharides (GUPS) were obtained with molecular weights of 1.06, 29.1, and 14.9 kDa, respectively. The monosaccharide compositions of GUPS-II and GUPS-III were similar, while that of GUPS-I was distinctively different. The results of scanning electron microscopy, FT-IR, and NMR suggested that GUPS-II and GUPS-III were flaky with a smooth surface and contained α- and β-glycosidic linkages, while GUPS-I was granulated and contained only α-glycosidic linkages. Moreover, GUPS-II and GUPS-III exhibited better bioactivities on the maturation and cytokine production of dendritic cells (DCs) in vitro than that of GUPS-I. An in vivo experiment showed that only GUPS-II significantly enhanced the maturation of DCs. These results indicate that GUPS-II has the potential to be used in combination with cancer immunotherapy to enhance the therapeutic effect.
