Structural characterization and immunomodulatory activity of a new polysaccharide isolated from the radix of Platycodon grandiflorum

Abstract: In this study, a novel polysaccharide fraction (PGPⅣ‐1‐a) was isolated from Platycodon grandiflorum, and its structure properties and immunomodulatory activity were investigated.

“…The alcohol extract of P. grandiflorum contains saponins, flavonoids, polysaccharides, polyphenol, and other components 34 . A neutral heteropolysaccharide isolated from P. grandiflorum significantly stimulated the proliferation of RAW264.7 cells in the range of 100 μg/mL to 800 μg/mL (P < 0.05), enhanced its phagocytic ability and increased in a dose-dependent manner 35 . Ethanol extract and fermented extract of P. grandiflorum 17 significantly reduced NO concentration in LPS-induced RAW 264.7 cells in a dose-dependent manner (P < 0.05) 36 .…”

“…Moreover, the signal peak at 3.70/76.07 ppm was attributed to the correlation of H-1/C-4, confirming the existence of the →4)-α-Glcp-(1→ residue, and the signal peaks at 4.10/103.98 ppm, 3.67/103.98 ppm, and 3.57/104.98 ppm were ascribed to the correlations of H-4/C-2, H-1/C-2, and H-6/C-2, respectively, validating the presence of the Fruf-(2→ and →6-)-β-Fruf-(2→ residues. By employing the same methodology and comparing it with previously published NMR data [ 13 , 14 , 15 , 16 , 17 , 18 , 19 ], the chemical shifts of other residues’ protons and carbons were sequentially deduced, as detailed in Table 3 . Based on these findings, a plausible structure for DT1-0 is proposed in Figure 9 .…”

“…In the cur- Macrophages are mononuclear phagocytes that initiate and engage in innate immunological responses, which play an important role in host defense by defending the infection of pathogens and foreign materials (Wang et al, 2020). The natural polysaccharides exert immunostimulatory effects by activating macrophage immune system functions, such as phagocytosis and cytokine secretion (Shan et al, 2020). The phagocytic ability is one of the most crucial parameters of macrophage activation.…”
