Gastrodia elata Blume polysaccharide (GEP) was extracted and then chemically characterised. Its antioxidant activity was evaluated in vitro and in vivo. The results of the in vitro investigation show that GEP consists of glucose with molecular weight of 875 185 Da and exhibits high hydroxyl radical scavenging, as well as 2,2-diphenyl-1picrylhydrazyl activity and reducing capacity. For antioxidant activity in vivo, D-galactose-induced-aged mice were orally administered with three different doses of GEP over a period of 6 weeks. The administration of GEP dosedependently increased the body weight gain rates, liver and brain indices, superoxide dismutase and glutathione peroxidase activities, and malondialdehyde levels in the sera and brains of ageing mice. These results suggest that GEP exhibits high antioxidant activity and can retard human ageing associated with free radicals.

A response surface and Box-Behnken design approach was applied to augment polysaccharide extraction from the residue of Rhizoma gastrodiae. Statistical analysis revealed that the linear and quadratic terms for three variables during extraction exhibited obvious effects on extraction yield. The optimum conditions were determined to be a liquid-to-solid ratio of 54 mL/g, an extraction temperature of 74 °C, an extraction time of 66 min, and three extractions. These conditions resulted in a maximum Rhizoma gastrodiae polysaccharide (RGP) extraction yield of 6.11% ± 0.13%. Two homogeneous polysaccharides (RGP-1a and RGP-1b) were obtained using DEAE cellulose-52 and Sephadex G-100 columns. The preliminary characterization of RGP-1a and RGP-1b was performed using HPLC-RID, HPGPC, and FTIR. Tests of the immunological activity in vitro showed that the two polysaccharides could significantly stimulate macrophages to release NO and enhance phagocytosis in a dose-dependent manner. In particular, RGP-1b (200 μg/mL) and LPS (2 μg/mL) had almost the same influence on the NO production and phagocytic activity of RAW 264.7 macrophages (p > 0.05). All the data obtained indicate that RGP-1a and RGP-1b have the potential to be developed as a health food.

BACKGROUND Cyclophosphamide (CTX) is a widely used chemotherapeutic agent for the treatment of malignant tumors and autoimmune diseases. However, it can cause immunosuppression and damage the intestinal mucosa. The development of new agents to counteract these side effects is becoming increasingly important. Previous studies have shown that the polysaccharides from Gastrodia elata (GEPs) have strong immune‐enhancing effects; however, their functions regarding the intestines and the underlying mechanism are still unclear. In this study, the effects of GEPs on immunomodulatory activity, intestinal barrier function, and gut microbiota regulation were investigated in a mouse model of CTX‐induced immunosuppression. RESULTS Gastrodia elata polysaccharides attenuated the CTX‐induced decrease in organ indices of the thymus and spleen, and promoted the secretion of immune‐related cytokines and immunoglobulins in the serum. They also improved the intestinal pathology and restored the intestinal barrier function by elevating the expression of intestinal tight junction proteins, occludin and ZO‐1. Moreover, GEPs restored the composition and abundance of the gut microbiota and increased the short‐chain fatty acid (SCFA) content in the colon. The abundance of SCFA‐producing bacteria (Muribaculaceae, Prevotellaceae, and Bacteroidaceae) also increased. CONCLUSIONS Gastrodia elata polysaccharides can effectively alleviate immunosuppression and regulate the intestinal barrier integrity and the structure of gut microbiota in CTX‐treated mice. They may be used as ingredients to develop functional foods for intestinal health. © 2023 Society of Chemical Industry.
