Melanoidins, formed at the final stages of Maillard reaction (MR), present important physiological activities, but their structure and reaction pathways are largely unknown in real food systems. In the present work, these bioactive compounds derived from Brewers' spent grain (BSG) with different roasting degrees were analysed. Results showed that BSG could be considered a more important source of bioavailable antioxidants than what it was reported in previous studies. During MR development, proteinic structures are partially lost while a carbohydrate skeleton is formed. The crystallinity is reduced and the structure sets into a conformation with a peak centred at 20°(2h), stable with the increasing thermal treatment. While MR continues the development of browning compounds increases, the antioxidant activity rises and phenolic compounds are bind to the melanoidin structure (P ≤ 0.05). Moreover, FTIR analysis showed a dominance of a-glyosidic linkage and the presence of furanose rings in the melanoidins structure.

In this study, an enzyme‐assisted extraction method was used to extract Orchis chusua D. Don (Salep) polysaccharide (SP), which was then modified by sulfation, acetylation, phosphorylation, and carboxymethylation to obtain modified polysaccharides. Furthermore, their degree of substitution, chemical composition, and molecular weight were evaluated. The primary structural features were characterized by UV spectra, FT‐IR spectra, Congo‐red test, and scanning electron microscope. The phosphorylated polysaccharide (SP‐P) was demonstrated the highest scavenging ability on hydroxyl radical and growth‐promoting activity on Lactobacillus Bulgaricus. The carboxymethylated polysaccharide (SP‐C) was exhibited the strongest DPPH and ABTS radical scavenging effects. The acetylated polysaccharide (SP‐A) displayed the best proliferation effects on Bifidobacterium adolescentis, whereas the sulfated polysaccharide (SP‐S) maintained moderately stable antioxidant and probiotic ability. These findings indicate that the modified polysaccharides had their potential significance as new antioxidants and probiotics for the food industry. Practical Application This article provides a new source for the development of polysaccharide derivatives as new antioxidants and probiotics for the food industry.

Treatment for tumors depends on host immune system. The antitumor and immunoregulatory activities of Ulva lactuca polysaccharide (ULP) were evaluated in H22 tumorbearing mice and cyclophosphamide-induced immunosuppressed mice, respectively.The structural properties of ULP were identified through multi-angle laser light scattering, high-performance liquid chromatography, Fourier-transformed infrared, and nuclear magnetic resonance. It was composed of -D-Manp-(1→, →2,4)--L-Rhap-(1→, -D-GlcpA-(1→, -GalpA-(1→, →2,4)--D-Glcp-(1→, and →6)--D-Galp-(1→ with the molecular weight of 1.46 × 10 5 Da. Its antioxidant, anti-inflammatory, and antitumor effects were determined. Liver and tumor tissues were collected for histopathological, immunohistochemical, and western blotting analysis. ULP enhanced the expression of p53 to inhibit tumorigenesis, promoted the activation of IKK , and inhibited the activation of p65 within the NF-B pathway. ULP inhibited the tumor growth through downregulating the expressions of PI3K/Akt and mTOR, and promoting BAX/Bcl-2 ratio. The inhibition of TRAF2/TNF-and CD31/VEGF achieved a direct killing effect on tumor cells and inhibited tumor proliferation by inhibiting angiogenesis, respectively. Moreover, ULP increased the levels of immunoglobulin M and total superoxide dismutase, decreased the level of methane dicarboxylic aldehyde, and inhibited the activation of PI3K/AKT/mTOR/p70S6k pathways.
