Structural Changes of Cell Wall Mannans of Candida guilliermondii IFO 10279 Strain Cells Cultured at High Temperature

Abstract: The morphology, structure, and antigenicity of the cells and the cell wall mannans of the Candida guilliermondii IFO 10279 strain cultivated at 33 and 34°C for 48 h in yeast extract-added Sabouraud liquid medium (YSLM) were compared with those cultivated at 27°C and 33°C and then at 27°C (33-27°C). This strain showed little growth at higher than 35°C. The density of the yeast formed cells decreased, with dry weights of about 50% at 33 and 34°C, and only the cells at 34°C revealed a failure of cytokinesis. The … Show more

“…We previously reported that CAWS727, prepared from the culture supernatant of C. albicans NBRC1385 cultured at 27ºC for 2 days in pH-controlled C-limiting medium (pH 7), did not induce vasculitis, and this was attributed to the presence of β-mannosyl linkages 11) . These findings were strongly supported by Shibata et al, who reported that βmannosyl transferase was only active at approximately 27ºC and neutral pH 57,58,61) . In the present study, we compared the structures of MN fractions using NMR analysis and found that signals assignable to β -mannosyl linkages were detected in various species, including C. albicans, C. dubliniensis, C. glabrata, C. guilliermondii, a n d C. tropicalis 16,27,28,36,37,42,57,58,60,[62][63][64][65] .…”

“…5, the spectra of MN fractions in each species were similar to each other and showed species-specific signal patterns. According to Shibata's assignment, the anomeric signals of α -mannose and βmannose residues were characteristic to species and appeared at approximately 5.6-4.9 and 5.0-4.7 27,42,[57][58][59] . Consistent with these results, all of the spectra contained signals assignable to α-mannan.…”

“…C. guilliermondii cell wall contains chitin, which increases in amount in response to exposure to sub-lethal concentrations of caspofungin (Walker et al, 2013). Furthermore, structural analysis of C. guilliermondii cell wall mannans indicated the presence of α1,2-, α1,3- and β1,2-mannose units, suggesting a similar organization to the C. albicans mannans (Okawa et al, 2006). …”

“…Regulation of beta-1,2-mannosyl linkages by culture condition is also reported in Candida guilliermondii. 23) From the data of immunochemistry and NMR spectra, CAWS 27-5.2 and CAWS 27-7.0 contained significantly higher concentrations of beta-1,2-linked mannooligosaccharides.…”
