Coronary Vasculitis Induced in Mice by Cell Wall Mannoprotein Fractions of Clinically Isolated Candida Species

Abstract: Kawasaki disease (KD) is an inflammatory disease that was identified by Professor Tomisaku Kawasaki in 1961. Candida albicans-derived substances (CADS) such as the hot water extract of C. albicans and Candida water-soluble fractions (CAWS) induce coronary vasculitis similar to KD in mice. An increasing proportion of deep-seated candidiasis cases are caused by nonalbicans Candida and are often resistant to antifungal drugs. We herein investigated whether the mannoprotein fractions (MN fractions) of clinically i… Show more

“…Following this, Candida sp. derived substances were demonstrated to induce coronary vasculitis in experimental mice models ( 64 , 65 ). This implies that the putative causative agent, possibly a fungal toxin, is transported across by strong air currents that develop in upper troposphere, supporting the theory that KD may be caused by an ubiquitous air-borne infectious organism.…”

“…Dectin-2 (CLEC6A) is a CLR that mainly exists in dendritic cells, macrophages, and neutrophils. Dectin-2 has been proven to recognize α-Mannan in the fungal cell walls (Figure 3) [59,60]. After α-mannan is recognized by Dectin-2, it can also send signals through CARD9, causing the activation of NF-κB, and inducing the production of inflammatory cytokines [61].…”

“…A wide variety of innate and adaptive immune cells, including monocytes, macrophages, neutrophils, and Tlymphocytes, infiltrate the coronary artery wall in KD, which is linked to the condition 3,[17][18][19][20][21][22] . These cells produce a large quantity of inflammatory cytokines such as Interleukin 1 (IL1), tumor necrosis factor a (TNF), and Interleukin 17 (IL17) that can directly disturb the vascular homeostasis [23][24][25] and lead to vascular dysfunction as described elsewhere [23][24][25][26] .…”
