Strontium ranelate-loaded PLGA porous microspheres enhancing the osteogenesis of MC3T3-E1 cells

Abstract: Biodegradable poly lactic-co-glycolic acid (PLGA) has been used as a tissue engineering scaffold as well as a carrier for the delivery of proteins, drugs, and other macromolecules.

“…[8][9][10][11] MSCs have been the focus of recent studies in bone tissue engineering, in which they are seeded in a scaffold and induced to generate new osteogenic cells via osteoinductive cues. [12][13][14] Appropriate bioactive cues encapsulated within the scaffolds can not only regulate cell adhesion and proliferation, but also enhance osteogenic differentiation. For example, incorporation of hydroxyapatite (HA) into the formulations of scaffolds enhanced the osteoinductivity 15,16 since bone is primarily composed of HA 17 .…”

“…There are some studies of porous scaffolds via mechanically mixing β-TCP/β-CaSi with different β-TCP mass fraction 17 , 19 21 but little of research on excellent osteogenic capability has been confirmed. On the other hand, granular form of bioceramics has an advantage of suiting for non-geometrically well-defined craniomaxillofacial bone defects accompanied with favorable interconnected macropore structures and is also potentially used as drug delivery systems 22 , 23 . Recently, we have developed a gradient hybrid design to fabricate core-shell β-TCP‒β-CaSi (CaP@CaSi, CaSi@CaP) microspheres conceiving respective biological performance to be exerted in specific layer 24 .…”

“…SrR, a classic anti-osteoporosis drug, has been gradually recognized to have an anti-osteoarthritis effect and the scope of its clinical application has gradually expanded in recent years. The role of SrR in promoting osteogenesis (18,(24)(25)(26), chondrogenesis (14,15,18,27) and angiogenesis (21,28) has been experimentally confirmed, but its anti-inflammatory Figure 1.
