Stretch–Activation of Angiotensin II Type 1 a Receptors Contributes to the Myogenic Response of Mouse Mesenteric and Renal Arteries

Abstract: Wall stretch is a major stimulus for the myogenic response of small arteries to pressure. Recent findings suggest that G protein‐coupled receptors can elicit a stretch response. Our aim was to determine if angiotensin II type 1 receptors (AT1R) in vascular smooth muscle cells (VSMC) exert mechanosensitivity and identify the downstream ion channel mediators of myogenic vasoconstriction. We used mice deficient in AT1R signaling molecules and putative ion channel targets, namely AT1R, angiotensinogen, TRPC6 chann… Show more

“…Mesenteric arteries were removed, quickly transferred to cold (4°C) oxygenated (95% O 2 /5% CO 2 ) physiological salt solution (PSS) (in mmol/l: 119 NaCl, 4.7 KCl, 1.2 KH 2 PO 4 , 25 NaHCO 3 , 1.2 Mg 2 SO 4 , 11.1 glucose, and 1.6 CaCl 2 ), and dissected into 2-mm rings (47). Each ring was positioned between 2 stainless steel wires (0.0394 mm diameter) in a 5-ml PSS-filled organ bath of small vessel myograph (DMT 610M, Danish Myo Technology).…”

“…KCNQ1 is mainly expressed in cardiac and epithelial cells, KCNQ2-5 are found in neuronal and primary sensory cells (1,4), and several KCNQ channels are also found in vascular smooth muscle (5)(6)(7). The neuronal KCNQ channels (KCNQ2-5) show properties of the M-current (8 -11), a slowly activating, non-inactivating outward rectifying potassium current that can be inhibited by muscarinic stimulation (12).…”

“…7 G-protein-coupled receptors like the angiotensin type 1 receptor possess mechanosensitive properties, independent of ligand binding, and may activate TRPs. 5,8 Recent studies also showed that interactions between SMCs and ECM are involved in MT. Pressureinduced deformation of ECM elements may activate integrins (eg, β1, β3) 9 and induces cytoskeletal remodeling and contraction through modulation of stretch-activated ion channel/ TRP and membrane depolarization.…”
