Stressors alter intercellular communication and exosome profile of nasopharyngeal carcinoma cells

Abstract: Our results suggest that the intercellular communication between tumor cells and surrounding stroma cells can be altered by microenvironment which increased quantity of exosomes and diversity of miRNAs in this study. Imbalance of oncogenic and tumor suppressor miRNAs caused by cytostatic treatments may influence the antiproliferative and metastasis inhibitory effect of cytostatic agents.

“…The presence and potential role of extracellular vesicles especially exosomes in follicular fluid has been reported in mare [ 25 ], women [ 68 ] and cow [ 22 , 69 ]. The quality and quantity of these extracellular vesicles vary depending on the physiological status of the cells, from where they are released [ 68 , 70 , 71 ] and are highly triggered by various stresses factors including diseases, heat stress as well as oxidative stress [ 72 ]. In agreement with that, our resultes demonstrated that culture media containing granulosa cells exposed to stress contained higher concentration of exosomes compared to those released by granulosa cells under normal conditions ( Fig 7 ).…”

“…Different kinds of exosomes have been isolated from the serum of NPC patients, such as EBV-related exosomes and pure nasopharyngeal carcinoma-derived exosomes 40 . These exosomes contain different contents and thus play differential roles in tumor-host crosstalk and modulate the immune response, angiogenesis, cell proliferation, cell-to-cell communication, and tumor invasion 41 , 42 .…”

“…Typically, 1 ml of culture medium will yield<1 µg of exosomal protein, while effective doses of exosomes range from 10 ug to100 ug. Certain techniques have been found to enhance exosome production, including the application of stressors such as low pH, hypoxia and anti-cancer drugs [109]. However, it is important to note that the effects of these techniques on exosome cargo are not fully understood, and caution must be taken in their evaluation.…”
