Streptococcus plasmid pAM alpha 1 is a composite of two separable replicons, one of which is closely related to Bacillus plasmid pBC16

Abstract: A tetracycline resistance plasmid of Streptococcus faecalis, pAMal, is shown to contain two independent sets of replication functions, separated from each other on either side by short (300to 400-base-pair) sequences of homology. The homologous sequences are oriented as direct repeats and therefore permit the dissociation of pAMal into its component replicons, referred to here as pAMalA1 and pAMalA2, as the reciprocal products of a simple intramolecular recombination. pAMalAl is a 4.6-kilobase plasmid which ca… Show more

“…The size of region III plus the size of region I was 4.82 + 0.36 kb, which is close to the size of pDL316 (4.7 kb) estimated from the TaqI digests by using a HaeIII-digested 4X174 size marker. The size of region III plus the size of region II was 4.6 ± 0.31 kb, which is virtually identical to the reported size of pAMa1o1 (18).…”

“…The faster-migrating fragment appeared to be the same size as the EcoRI fragment H of pJH1. A fragment of this size was also present in EcoRI-digested pAMoqA1 (lane 2), a small, 4.6-kb tetracycline resistance plasmid described recently by Perkins and Youngman (18). The larger EcoRI fragment from pDL316 was not correlated with any of the fragments from pJH1 and was slightly larger than the corresponding fragment from pAMaAj1.…”

“…2). These direct GGGC repeats and the multiple inverted, more or less perfect repeats including part of the GGGC sequences may be subject to site-specific breakage and joining, making this fragment available as a cassette to pick up antibiotic resistance genes and thus to trigger their transfer between gram-positive species via conjugative mobilization and/or interplasmid recombination (10,18,(26)(27)(28). The RepB and pIP1714 Pre(Mob) proteins are similar to several other plasmid-encoded proteins present in the data bank, but the corresponding genes are not necessarily linked on the same plasmid.…”

“…Intramolecular recombination at homologous sequences dissociates plasmid pAMctl, a tetracycline resistance plasmid of S. faecalis, into two circular molecules, each containing a single homologous sequence (16). Intermolecular recombination at homologous sequences may generate multiple-copy plasmids.…”
