Streptococcus lutetiensis Induces Autophagy via Oxidative Stress in Bovine Mammary Epithelial Cells

Abstract: Streptococcus lutetiensis, an emerging pathogen causing bovine mastitis, has not been well characterized. We reported that S. lutetiensis was pathogenic both in vivo and in vitro and caused inflammatory reactions in the mammary gland. However, roles of autophagy and oxidative stress in the pathogenesis of S. lutetiensis-induced mastitis are unclear. In this study, an autophagy model of S. lutetiensis-infected bovine mammary epithelial cells (bMECs) was used to assess oxidative stress and autophagy flux. Expres… Show more

“…4 b). To further investigate the protective effect of huc-MSCs-derived exosomes on autophagy, TEM was used to observe changes in autophagosomes [ 34 ]. Our results showed that the amount of autophagosome increased in BV2 cells pretreated with 100 mg/mL huc-MSCs-derived exosomes (Additional file 1 : Fig.…”

“…S. dysgalactiae can not only form biofilm but also invade and survive within mammary epithelial cells, and persist for long periods, which can avoid the bactericidal action of antibiotics, resulting in persistent infection ( Calvinho and Oliver, 1998 ). Streptococcus also can induce autophagy to increase inflammatory reactions of bovine mammary epithelial cells ( Chen et al, 2022 ; Qi et al, 2022 ). Because of the low intracellular accumulation and special intracellular environmental effects, it is difficult for the antibiotics to play a good bactericidal effect in intracellular.…”

“…Yang et al found that phosphorylated p62 activation enhances Nrf2 nuclear translocation and reduces ROS production and mitochondrial dysfunction [ 160 ]. Chen et al found that the crosstalk between oxidative stress and autophagy activity affects autophagy flux and inhibits downstream autophagy activity, and that the Nrf2-Keap-p62 signaling pathway is involved in interfering with normal cell functions [ 161 ]. In addition to p62, the autophagy adaptor protein NDP52 has been confirmed to participate in the crosstalk between the Nrf2 pathway and autophagy.…”
