In the present study, the discrimination of phytochemical content of Myrtus communis berries from different geographical origin and cultivars was explored by Liquid Chromatography-Electrospray Ionization-Fourier Transform-Mass Spectrometry (LC-ESI-FT-MS) metabolic profiling and quantitative analysis. Experiments were carried on myrtle plants grown in an experimental area of Sardinia region, obtained by the germination of seeds taken from berries collected in each part of the region. A preliminary untargeted approach on fruit's extracts was realized by collecting LC-ESI-FT-(Orbitrap)-MS data obtained by operating in negative ion mode and performing principal component analysis with the result of differentiation of samples. In a second step, targeted analysis with a reduced number of variables was realized. A data matrix was obtained by the data fusion of positive and negative ionization LC-ESI-MS results, by using as variables the peak areas of each known compounds. By the observation of principal component analysis, results found that anthocyanins, and mainly derivatives of cyanidin, are the principal marker compounds responsive for the discrimination of samples based on the geographical origin of the seeds. Based on this finding, finally, an LC-diode array detector method was developed, validated and applied for the quantitative analysis of berries' extracts based on 11 commercial standard compounds corresponding to the identified markers. Stevia rebaudiana Bertoni is a perennial medicinal herb indigenous to tropical regions of South America. The present study evaluated some physiological changes of the plant under cold in the absence and the presence of polyamine conditions. The results showed photosynthetic pigments and photochemical efficiency of PSII reduced in cold treatment conditions than control, significantly. However, a considerable increase in the parameters was observed in all of polyamine treated goups. Leaf compatible solutes such as proline and glycine betaine that were not shown significantly increasing at cold treatment groups, elevated in the 2 days cold-polyamine-treated group. The similar results were seen for free amino acid and total protein contents of the treated plants. It was also revealed that specific activity of some antioxidant enzymes such as catalase and ascorbate peroxidase increased in cold-treated or cold + polyamine-treated groups than related controls. It caused a significant decrease in H 2 O 2 and malondialdehyde contents of the treated plants. The results indicated an increasing in saturated long chain fatty acids after 4 days of cold at 4 °C. It was concluded that the polyamine supplement can induced a considerable tolerance in stevia plants at cold condition.

To optimise steviol glycoside analysis, several round-robin tests were organised by the European Stevia Association (EUSTAS). Seventeen laboratories participated in the testing. Only 8 laboratories have sent their results. In the first round-robin testing, 2 samples were analysed. The first sample had a purity of 96.2%. The second sample was a 4/5 dilution with NaHCO 3 of sample 1. This way, the drying process itself could be checked. The purity of sample 2 was 82.35%. The reported purities of sample 1 varied between 79.8 and 96.2%, those of sample 2 varied between 58.1 and 81.8%. The drying of sample 2 showed that weight loss was between 4.9 and 12.7%, demonstrating that not all laboratories dried the sample to a constant weight. In a second round-robin testing, the purity of the sample was 91.1%. The reported purities of the sample varied between 82.74 and 95.86%. About 3% purified RebB was added to the sample to check the quality of the analysis of this compound possessing a carboxylic group. The samples contained the following steviol glycosides: Reb D, Reb E, Reb A, ST, Reb F, Reb C, Dul A, Reb G, Rub, Reb B, SB and SM (1 lab). No SV was detected. The number of SVgly analysed in the different laboratories varied between 4 and 11. One lab only analysed ST and Reb A and gave a percentage composition of these compounds. To improve the accuracy of analysis, different suggestions are made, such as controlling the drying process of samples and standards, purity of standards, injection of sufficient material and use of solvent gradients to shorten the run time and to reduce integration errors. The results of this second round-robin tests are better than those of the first one.
