STC-1 Cells

Abstract: OriginSTC-1 was originally established from cells present in murine enteroendocrine tumours. These tumours arose in the duodenum of double transgenic mice expressing the rat insulin promoter linked to the simian virus 40 large T antigen and to the

“…STC-1 cells were derived from the duodenum of transgenic mice that expressed SV40 large T antigen downstream from a rat insulin promoter (36); they are widely accepted as a model of native EECs (37). STC-1 cells express several gastrointestinal hormones, such as cholecystokinin (CCK) and peptide YY (PYY), whose secretion is regulated in a manner similar to native EECs (38)(39)(40).…”

“…GLP-1 receptor agonists and inhibitors preventing GLP-1 degradation, for instance, are currently used in the clinic to treat patients with T2DM (Green & Flatt, 2007). There is a demand for robust enteroendocrine cell models for the routine screening of nutrients and secretagogues (McCarthy et al 2015). Established cell lines used for this purpose include: GLUtag -derived from an intestinal endocrine tumour arising in the large bowel in proglucagon-simian virus 40 large T antigen transgenic mice (Brubaker et al, 1998); NCI-H716 -a human cell line derived from a poorly differentiated adenocarcinoma of the caecum (De Bruine et al, 1992), and STC-1 which is a neuroendocrine cell line derived from tumours arising in transgenic mice (McCarthy et al 2015).…”

“…STC-1 cells originate from enteroendocrine tumors in the duodenum of double transgenic mice. (24) This cell line was used for all experiments. STC-1 cells were cultured (95% O 2 ; 5% CO 2 ; 37°C) in Dulbecco’s modified Eagle’s Medium (DMEM) containing 4.5 g/L D-glucose, 4 mM L-Glutamine (Sigma), supplemented with 10% FBS (Sigma), 100 U/mL penicillin and 100 mg/L streptomycin (ThermoFisher; DMEM+/+).…”
