SSCP polymorphism within a promoter of the bovine alpha S1 casein gene

Abstract: The detection of a new SSCP polymorphism within the promoter of the bovine alpha SI casein gene is described. The fragment of alpha SI casein gene promoter (218 bp) was PCR-amplified and then subjected to electrophoresis allowing the detection of SSCP polymorphism. Among 118 randomly chosen cows (Black-and-White) and 124 bulls (Black-and-White), we found two SSCP patterns, labelled R2 and R3, with respective frequencies of 0.05 and 0.95. The detected SSCP polymorphism may be useful in searching for association… Show more

“…One portion of semen was used to isolate genomic DNA by the MasterPure DNA Purification Kit (Epicentre). DNA was used to amplify 6 PCR products, namely: a 208 bp fragment of LGB promoter, designated LGB-R (Kaminski and Zabolewicz, 1998), a 240 bp fragment of LGB exon IV, designated LGB (Kaminski and Zabolewicz, 1997), a 214 bp fragment of CASK promoter, designated CASK-R (Kaminski, 1996), a 273 bp fragment of CASK exon IV-intron IV, designated CASK (Kaminski, 1998), a 483 bp fragment of LALBA 5' flanking region, designated LALBA (Kaminski, 1999) and a 218 bp fragment of CSN1S1 promoter, designated CSN1S1 (Kaminski and Jozefowicz, 2000). The breeding values of bulls were collected from the official ranking list of proven bulls published bi-annually by the National Animal Breeding Centre in Warsaw (Poland).…”

“…Polymorphism within the 5'-flanking region of the bovine b-lactoglobulin gene (b-lgPTU) was studied by Kamin u ski and Zabolewicz [46] in the Polish Black-andWhite dairy cattle, in which the two predominant b-lactoglobulin variants are also A and B. Moreover, within the promoter region (b-lgP) of the gene, several point mutations were found by SSCP analysis.…”
