Spermidine‐enhanced autophagic flux improves cardiac dysfunction following myocardial infarction by targeting the AMPK/mTOR signalling pathway

Abstract: Background and Purpose Spermidine, a natural polyamine, is abundant in mammalian cells and is involved in cell growth, proliferation, and regeneration. Recently, oral spermidine supplements were cardioprotective in age‐related cardiac dysfunction, through enhancing autophagic flux. However, the effect of spermidine on myocardial injury and cardiac dysfunction following myocardial infarction (MI) remains unknown. Experimental Approach We determined the effects of spermidine in a model of MI, Sprague–Dawley rats… Show more

“…180 Similarly, luteolin was found to enhance autophagic flux, improve mitochondria function, and inhibit apoptosis through Mst1 inhibition. 181 Aside from trehalose, melatonin, and luteolin, restoration of autophagic flux was also induced by nobiletin 182 and spermidine 183 in animal models of MI or MI-induced heart failure, and most of them exert protection against MI via activating autophagy or accelerating autophagy flux. “…Recently, it has been reported that autophagy inhibition or autophagy flux impairment can cause cardiac hypertrophy [3][4][5][6]. Enhanced autophagy may attenuate cardiac hypertrophy [7][8][9]. In bronchial epithelial cells, nicotine was reported to be involved in the regulation of autophagy and emphysema progression [10,11].…”

“…Recent reports have pointed at the ability of SPD to counteract disc degeneration via enhancement of autophagy in the cells of the nucleus pulposus [9] as well as its ability to reduce oxidative damage and inflammation in a model of cardiac infarction via enhancing the AMPK/ mTOR-mediated autophagic flux [10].…”
