Spectroscopic investigations of the molecular interaction of anticancer drug mitoxantrone with non-ionic surfactant micelles

Abstract: These results on simple model systems miming the drug-membrane interactions contribute to the elucidation of the behaviour of the drug in vivo, as well as the possible utilization of surfactant micelles as drug carriers.

“…As shown by light and electron microscopy, the intravenous or intra-arterial administration of octoxynol-9 selectively injures the blood vessel endothelial cell (McLeod and Piper, 1992b, Bourreau et al, 1993). Octoxynol-9 is used both in vivo (Pridgen et al, 2011, Enanche and Volanschi, 2012) and in vitro (Eddy et al, 2012, Murphy et al, 2012, Soni et al, 2012). While new to the field of pain research, octoxynol-9 has been used for experimental intervention in cardiovascular studies for many years (Connor et al, 1989, Sarvazyan, 1998, Mink et al, 2007).…”

“…The surfactant concentration values at which detectable change in the spectral parameters have been evidenced was slightly higher than the cmc tensiometrically determinate. This result might be ascribed to the shift of the micellization process due to the presence of an hydrophobic molecules [4,10]. Therefore, the cmc values determined from the change in the absorption spectrum of NR aqueous solution have been considered in the calculations.…”

“…Classically, the binding constant of a probe [4,5], being different hydrophilicity, to micellar aggregates is estimated applying the two-pseudo phases model by considering only the change in the absorbance at the k max as a function of surfactant concentration. This situation mainly represents the palisade layer of the micelles where the solubilisation of the probe is governed by both the hydrophilic and hydrophobic interactions.…”

“…Surfactant micelles are found useful to modulate drug absorption as well as to protect the active drug molecules from the adverse environmental conditions (e.g. gastric acid) [8]. Besides the solubilizing capacity, the hydrophilic corona of micelle is capable to create a highly water-bound barrier, which potentially blocks the adhesion of other hydrophobic compounds [9][10][11].…”
