Spectrophotometric Determination of Protein with DBC-Chlorophosphonazo

Abstract: A new spectrophotometric method for the determination of protein has been developed using the property that bovine serum albumin and DBC-chlorophosphonazo form a compound. The maximum absorption wavelength of the compound is 606 nm. Beer's law is obeyed over the range of 25-250 µg/mL of bovine serum albumin contents. The detection limit of this method is 9.52 µg/mL. Protein content in human serum has been successfully determined.

“…To one of the volumetric flasks, 2-7 mg of the protein standard solution was added and diluted with water to the mark and shaken. The solution was allowed to stand for 35 min at room temperature and using the corresponding reagent blank as a reference and with 1 cm cell absorbance was measured at 600 nm 16 .…”

“…An ASAP 2020 surface and porosity analyzer (Micromeritics Instrument Corporation, USA) was used to determine the N2 adsorption-desorption isotherm curve of sample at 77 K. By means of BET (Brunauer-Emmett-Teller) method 13 and BJH (Barrett-Joyner-Halenda) method 14 , the specific surface area and average pore size of sample were calculate, respectively. α-Amylase component content determination in (SBA-15)-(α-amylase) was obtained by the spectrophotometry 11 with subtraction method with a 722S type spectrophotometer (Shanghai Lengguang Technology Co., Ltd., China).…”
