Spectrophotometric Determination of Gentamicin Loaded PLGA Microparticles and Method Validation via Ninhydrin-Gentamicin Complex As A Rapid Quantification Approach

Abstract: The purpose of this study was to provide a quantification method with rapid, sensitive, reproducible and cost effective for gentamicin in the form of ninhydrin-gentamicin complex. The utilization of spectrophotometric module to validate the method development for gentamicin loaded microparticles was intended to provide alternative quantification method without undermining the sensitivity of the developed method. The microparticles fabrication process was proven to be suitable in encapsulating gentamicin by usi… Show more

“…Concentrations of gentamicin in the eluted samples were analyzed through the ninhydrin colorimetric assay, which has been proved to be a reproducible, efficient, and safe method of detecting concentrations of gentamicin (45,46). Briefly, 5 mg/ml ninhydrin stock solution was formed by dissolving ninhydrin powder (VWR International, Radnor, PA) in PBS.…”

“…Amoxicillin was determined based on its colored condensation adduct with ascorbic acid absorbing at λ max = 410 nm, according to the procedure described by El-Shafie et al (limit of detection of 6 ± 0.1 μg/mL) [ 36 ]. Gentamicin sulfate was determined according to the method described by Ismail et al, based on the absorbance maximum λ max = 418 nm of gentamicin: ninhydrin complexes (limit of detection of 16 ± 0.3 μg/mL) [ 37 ].…”

“…Spectrophotometric method was developed via ninhydrin-Gentamicin complex as a rapid quantification approach for the determination of Gentamicin loaded with poly (lactic-co-glycolic acid) PLGA microparticles that was suitable for Gentamicin encapsulation without compromising the efficacy of the antibiotic itself [137] .…”
