SPECT-computed tomography in rats with TNBS-induced colitis: A first step toward functional imaging

Abstract: AIMTo assess the feasibility of SPECT-computed tomography (CT) in rats with trinitrobenzene sulfonic acid (TNBS)-induced acute colitis and confront it with model inflammatory characteristics.METHODSColitis was induced in Sprague-Dawley rats by intrarectal injection of TNBS (n = 10) while controls received vehicle (n = 10). SPECT-CT with intravenous injection of 10 MBq of 67Ga-Citrate was performed at day 2. SPECT-CT criteria were colon wall thickness and maximal wall signal intensity. Laboratory parameters wer… Show more

“…Relatively to the bowel thickness ( n = 9), it is expected an increase in the thickness of the gut wall in the presence of inflammation. Maximal thickness, correlated to colon cyclooxygenase (COX-2) expression, is significantly increased in TNBS groups compared to the control groups [102]. COX-2 is an inflammatory marker involved in angiogenesis processes and chronic inflammation and be more relevant in chronic colitis assessment [103].…”

“…Moreover, a commercially available WMC is too large for use in rodents. Scintigraphy is also used to measure SI transit but requires a specialized facility and exposure to radiation [2, 10, 16]. Abdominal ultrasonography, however, provides noninvasive, real-time images and measures SI motility quickly.…”

“…Macroscopic score evaluation of the colon was based on descriptions by Morris et al (1989) [ 145 ]. Colon length ( n = 28) is evaluated in our studies as a picture of intestinal damage and measured as a marker of tissue integrity, which is determined using a measuring scale; the literature points to a reduction in colonic length after TNBS treatment, indicating a reduction in the colon length after the disease installation [ 174 , 175 , 176 , 177 ].…”
