Specificity of human antibodies to oncovirus glycoproteins: recognition of antigen by natural antibodies directed against carbohydrate structures.

Abstract: Antibodies in human sera from healthy individuals were shown to be reactive with highly purified 70,00OA-totn envelope glycoprotein (gp70) of the simian sarcoma virus-simian sarcoma-associated virus (SSV-SSAV) complex in radioimmunoprecipitation assays under certain conditions. The specificity of the reaction was analyzed in absorption tests with normal human serum proteins, assays of viral gp7O antigenicity after exposure to exo-and endoglycosidases or trypsin, and carbohydrate hapten inhibition studies. On t… Show more

“…However, the epitopes responsible have been characterized in relatively few studies, and in some of the older reports, this seroreactivity turned out to be to carbohydrate antigens present on the viruses due to their production in nonhuman cell lines (31,455). Antibodies to HERV proteins have also been described, particularly in patients with testicular tumors (60,430) and melanoma (82), but such antibodies can also be present in healthy individuals (24,208).…”

“…Such techniques cannot accurately distinguish specific and nonspecific reactions of sera. Some authors believe that most reactiveness of human sera against viral proteins could be accounted for by heterophilic antibodies that bind the carbohydrate moieties of viral glycoproteins (Barbacid et al, 1980;Snyder & Fleissner, 1980). Nevertheless, human sera have been reported to react with core MMTV proteins using radioimmunoprecipitation (RIP) and ELISA with purified MMTV proteins (Zotter et al, 1983;Dion et al, 1987).…”
