Species differences in the pharmacokinetics of recainam, a new anti‐arrhythmic drug

Abstract: The pharmacokinetics of recainam, an anti-arrhythmic drug, were compared in mice, rats, rabbits, dogs, rhesus monkeys, and man. Bioavailability was virtually complete in monkeys and dogs, 67 per cent in man and 51 per cent in rats. Non-linear kinetics between the oral and i.v. dose in rabbits precluded estimation of bioavailability. Linear plasma dose proportionality occurred in dogs between 6 and 60 mg kg-1 oral doses and rhesus monkeys between 1 and 15 mg kg-1 i.v. doses. A greater than proportional increase… Show more

“…CL PO data for rats ( CL PO,rat ; range, 0.300–27778 mL/min/kg), dogs ( CL PO,dog ; range, 0.150–1598 mL/min/kg), and humans ( CL PO,man ; range, 0.150–3767 mL/min/kg) after oral administration of 87 drugs were obtained from the literature 6–128. The CL PO data for other species (mice, rabbits, monkeys, baboons, or guinea pigs) were also obtained to apply allometric approaches.…”
