Speciation of Organic and Inorganic Selenium in Selenium-enriched Eggs by Hydride Generation Atomic Fluorescence Spectrometry

Abstract: A new method was established for the speciation of Se by hydride generation-atomic fluorescence spectrometry. Effective separation of organic and inorganic Se in Se-enriched eggs was achieved by precipitating albumen with trichloroacetic acid. The detection limit for Se was 0.07 μg L −1 . The proposed method was successfully applied for the determination of organic and inorganic Se as well as total Se in Se-enriched eggs with the recovery of 90.1-112% and the relative standard deviation (n=5) of 0.6-3.4%. Se i… Show more

“…The concentrations of SeCys 2 and SeMet were 81.80 ± 0.53 µg kg −1 and 6.96 ± 0.20 µg kg −1 (as Se), respectively. According to reports, the intake of Se converted by hen organism was bonded to protein and present mainly in organic forms in eggs [13,14]. The results of Se species extracted from egg by enzymatic hydrolysis were consistent with this conclusion.…”

“…The detection limits were 60, 3, and 10 µg kg −1 (dry weight) for SeCys, Se(IV), and SeMet, respectively, and the precision was 5-10% [13]. Sun and Feng developed a method using HG-AFS by depositing albumen with trichloroacetic acid to determination organic and inorganic Se in fresh Se-enriched eggs [14]. Up to now, there were few reports about the analysis method of Se species in fresh liquid whole egg.…”

“…(2005a) so ugotavljali tudi vsebnost selena v ribah, kjer ga je 15,3-68,6 µg/100 g, v piščančjem mesu 9,7-15,4 µg/100 g) in v puranjem mesu 9,9-11,6 µg/100 g. Jajca v povprečju vsebujejo 26 µg selena v 100 g vzorca. Z dodatkom selena h krmni mešanici za kokoši je možno vsebnost selenometionina in selenocisteina v jajcih močno povečati (Sun in Feng, 2011).…”

“…Heras et al 22 reviewed the diverse analytical techniques employed to elucidate the structures of Se-containing proteins and presented the best characterized proteins, highlighting the many challenges involved. The relative proportion of organic Se and iSe in enriched eggs was determined using a new HG-AFS method developed by Sun and Feng 175 giving an LOD of 0.07 mg L À1 . The amount of organic Se was 3 to 7 times that of iSe.…”
