Spatial learning impairment in a murine model of AIDS

Abstract: Mice infected with an immunosuppressive murine leukemia virus (MuLV) mixture, LP-BM5, displayed profound and selective deficits in spatial learning in a modified Morris water maze. These deficits appeared before the appearance of gross neurological impairment or histopathological changes in the central nervous system. Thus, LP-BM5-infected mice displayed deficits in several aspects of trained performance compared to controls. Furthermore, a failure to exhibit any evidence of task acquisition in this maze was o… Show more

“…Based on previous experiments of Sei et al (1992), the performance of animals in reaching the target (target latency) was defined as 'good' (≤40 s), 'average' (41-90 s) and 'poor' (> 90 s). Animals were trained once daily beginning at 3 p.m.…”

“…In mice with MAIDS, pancreatic lesions of the exocrine system with a predominance of infiltrating Th2 cells have been described (28); moreover, CNS damage and cognitive deficits develop concurrently with the immunodeficiency (29)(30)(31). Hence, the thiol content in the pancreas and brain was also analyzed.…”

“…Moreover, it has been reported that LP-BM5 infection is accompanied by a pancreatitis-like injury (28) and by central nervous system (CNS) damage (29)(30)(31). Hence, to investigate whether the thiol content could play a role in the alterations of these organs, GSH and cysteine levels were also measured in the pancreas and brain.…”

“…The open-field water maze or 'Morris water maze' has been used extensively to examine behavioural, neural and physiological substrates of spatial mapping and learning in rodents (Morris, 1984). Several studies, utilising a range of infectious agents including parasitic nematodes (Kavaliers and Colwell, 1995;Braithwaite et al, 1998) and immune deficiency virus (Sei et al, 1992) (Gundra et al, 2011). The wide availability of knockout mice would also allow for direct correlation between specific cytokines and behavioural alterations.…”
