Soybean greatly reduces valproic acid plasma concentrations: A food–drug interaction study

Abstract: The aim of this study was to investigate the effects of soy on the pharmacokinetics and pharmacodynamics of valproic acid (VPA). In a preclinical study, rats were pretreated with two different amounts of soy extract for five days (150 mg/kg and 500 mg/kg), which resulted in decreases of 57% and 65% in the Cmax of VPA, respectively. AUC of VPA decreased to 83% and 70% in the soy pretreatment groups. Interestingly, the excretion rate of VPA glucuronide (VPAG) was higher in the soy-fed groups. Levels of UDP-glucu… Show more

“…VPA C max values were decreased to 57% for those pretreated with 150 mg soy, and to 65% for those treated with 500 mg. Of note, gamma aminobutyric acid (GABA) concentrations in the brain were increased within 5 min after VPA injection. In the soy-treated group, concentrations of GABA in the brain were significantly inhibited throughout the entire study period; however, plasma levels of GABA were not significantly different between the treatment and control groups (Marahatta et al 2014).…”

“…Limitations of the study include: (1) the sample size was relatively small (137 patients); (2) despite their influence on VPA metabolism, many variants were not selected due to low frequency in the population (MAF < 0.05) [7,20]; (3) possible influence of food-drug interactions on VPA plasma concentration was not evaluated [18].…”

“…Additionally, fooddrug interactions also influence the pharmacokinetics of VPA. For example, soybean intake can greatly decrease the plasma concentration of VPA [18].…”
