“…Because f3I1 spectrin cleavage at approximately 10 micromolar levels of Ca2+ occurs only in the presence of active CaM and is kinetically slower (185), spectrin together with CaM and micro-calpain conceptually form a system that: (1) in the absence of calcium, constituitively cross-links actin filaments; (2) when activated by calpain cleavage, exhibits reversible Ca 2 + -regulated actin cross-linking; and (3) with sustained Ca 2 + and calpain activity, irreversibly disassembles. A now considerable body of evidence indicates that the calpain cleavage of a II spectrin at tyr 1177 is a common sequel of many physiologic and pathologic processes, from NMDA receptor stimulation to secretion (reviewed in 165). The significance of these observations vis-a-vis receptor organization or function remains to be elucidated.…”
