SOMATIC EMBRYOGENESIS OF CARROT IN HORMONE‐FREE MEDIUM: EXTERNAL pH CONTROL OVER MORPHOGENESIS

Abstract: Cultures of preglobular stage proembryos (PGSPs) were initiated from mechanically wounded mature zygotic embryos of carrot, Daucus carota, on a hormone-free, semisolid medium. These PGSPs have been maintained and multiplied for extended periods without their progression into later embryo stages on the same hormone-free medium containing 1 mM NH4+ as the sole nitrogen source. Sustained maintenance of cultures comprised exclusively of PGSPs was dependent on medium pH throughout the culture period. Best growth… Show more

“…When we used MS medium supplemented with 2,4 and BAP (Table 1 ; original MS Teheran medium) callus and embryogenic callus induction, and subsequent shoots regeneration for all explants tested from both locations was lower. This medium was sterilized by autoclaving what can affect the final quality of induction media [ 83 ] including pH what can have an impact on the cellular morphology of cultures and regeneration capacity as well [ 84 , 85 ].…”

“…Ammonium (NH 4 + ) provided as the sole source of nitrogen sustains growth in many plant species (Britto andKronzucker 2002, Ali et al 2013). However, when supplied at a stressful, but not harmful concentration, ammonium may alter cell differentiation and trigger somatic embryogenesis induction (Smith andKrikorian 1990, Leljak-Levanić et al 2004). The effects of ammonium assimilation on embryogenic carrot cell multiplication without development into later embryo stages have been attributed mainly to culture medium acidification (Smith and Krikorian 1992).…”
