Somatic embryogenesis in Araucaria angustifolia

Abstract: Immature and mature zygotic embryos were used as source of explants for induction of somatic embryogenesis in Araucaria angustifolia. Embryogenic cultures (EC) were only obtained from immature zygotic embryos. Basic medium, carbon source, and genotype showed a significant influence on the formation of stage I somatic embryos (SE). When EC were submitted to maturation conditions, SE continued their individual development until stage II, but mature embryos were not obtained. Proteins secreted by embryogenic cult… Show more

“…Here, we investigated the role of both NSCs and the structural carbohydrates of the cell wall during A . angustifolia embryogenesis, comparing a cell line that was responsive to ABA, a blocked cell line, and the globular stage of zygotic embryogenesis, since the two cell lines were induced from immature zygotic embryos [ 84 ].…”

“…As we also report in our system here, promotivemitogenic factors are released in conditioned medium. 64 For example, phytosulfokine a, 21,22 cell wall remodeling enzymes, [65][66][67][68] and parietal fragments such as AGPs, [31][32][33][34][35][36][37]69 or oligogalacturonide pectic fragments have been reported. 68 The use of CM obtained from wild type carrot cell suspension favored embryo development in the thermosensitive mutant ts11 that normally produced somatic embryo arrested at the globular stage.…”

“…Essas moléculas hidrolisam eficientemente a quitina, presente no citoesqueleto da hifa fúngica (Ponstein et al, 1994).
