Somatic Embryogenesis and Plant Regeneration from in-vitro-grown Leaf Explants of Rose

Abstract: Somatic embryogenesis was initiated from in vitro-grown leaf explants of rose using an induction period of 4 weeks on MS basal medium supplemented with auxin followed by several subcultures on MS basal medium with cytokinin. `4th of July' showed the highest regeneration frequency (24.4%) on 5.3 μm NAA followed by culture on medium containing 18.2 μm zeatin. `Tournament of Roses' produced somatic embryos when cultured for 4 weeks on medium conta… Show more

“…Despite the fact that somatic embryogenesis from explants grown and maintained in vitro was successfully reported in many plant species (e.g., rhizomatous Iris (Laublin et al 1991), Camellia sinensis O. Kunze (Kato 1996), Rosa hybrida and Rosa multiflora (Kim et al 2004), Pyrus communis L. (Javadi et al 2013) and Olea europaea L. ), this approach has been scarcely studied in date palm (Al-Mayahi 2015). Therefore, the aim of this investigation was to develop a reproducible somatic embryogenesis pathway from buds and shoots maintained in vitro of date palm cv.…”

“…There are several reports on adventitious shoot regeneration and somatic embryo formation from different explants of roses (Tweddle et al 1984;Lloyd et al 1988;Burger et al 1990;Noriega and Sondahl 1991;Kunitake et al 1993;Arene et al 1993;Firoozabady et al 1994;Rosu et al 1995;Hsia and Korban 1996;Kim et al 2003;Pati et al 2004), but most protocols are useful only for specific genotypes, or occur at such low frequencies thus limiting value of these protocols for most roses. Fortunately, researchers also succeeded in obtaining pure forms from chimeras by rapid proliferation in vitro (Hall et al 1986;McPheeters and Skirvin 1989;Rosu et al 1995).…”

“…chinensis cv. Old Blush had previously been reported as producing non-embryogenic callus after induction of foliar explants with 4.5 lM 2,4-D hormone (Kim et al 2003). Here, the response of Old Blush callus to somatic embryogenic induction was triggered by replacing 2,4-D with 8.06 lM NAA.…”
