Somatic embryogenesis and plant regeneration in Cichorium intybus L. (Witloof, Compositae)

Abstract: Somatic embryogenesis of Cichorium intybus L. var. 'Carolus' is induced using cubical pieces of mature tap roots with an intervening callus phase. A Murashige and Skoog's (MS) semi solid basal medium supplemented with 2,4-dichlorophenoxyacetic acid (0.02 or 0.2 mg/l) and benzylaminopurine (0.25 mg/l) and a liquid MS medium devoid of growth regulators are used respectively for induction of callus and somatic embryoids and for further development and germination. Regeneration from the nodular proembryonal stage … Show more

“…It has been reported that higher concentrations (6%) of sucrose increases somatic embryo maturation in Picea mariana (Trembly and Trembly 1995). There have been several reports of increased somatic embryogenesis on medium containing reduced nutrients (Heirwegh et al 1985;Smith and Krikorian 1989;Nadel et al 1990;Choi and Soh 1996). In the present study we found that media dilution reduces embryo maturation and MS medium dilution up to 50% significantly improved germination.…”

“…Cytokinins have been found useful in fostering somatic embryo maturation (Fujimura & Komamine 1975), and cotyledon development (Ammirato & Steward 1971). The use of 2,4-D and BA for initiation of somatic embryogenesis has been reported for other species of Asteraceae (Heirwegh et al 1985;May & Trigiano 1991). However, in this study, development beyond the globular stage was inhibited by remaining on medium containing these plant growth regulators.…”

“…However, Staden and Bennett (1991) reported that the tissue from galls induced by Procecidochares utilis on the stems of croftonweed produce callus on a cytokinin-free medium, which resulted from much higher levels of endogenous cytokinin in gall tissue than that in normal stem tissue. In previous reports, (2,4-dichlorophenoxy)acetic acid (2,4-D), alone or combined with BA, was used to induce callus (Chengalrayan and Gallo-Meagher 2001;Ferreira and Handro 1988;Heirwegh et al 1985;Xu and Jia 1996). Different cytokinin and auxin combinations and concentrations had a significant (P , 0.01) effect on callus induction (Tables 1-3).…”
