Solid support resins and affinity purification mass spectrometry

Abstract: Co-affinity purification-mass spectrometry (CoAP-MS) is a primary technology for elucidating the protein-protein interactions that form the basis of all biological processes. A critical component of CoAP-MS is the affinity purification (AP) of the bait protein, usually by immobilization of an antibody to a solid-phase resin. This Minireview discusses common resins, reagents, tagging methods, and their consideration for successful AP of tagged proteins. We discuss our experiences with different solid supports, … Show more

“…Further procedures can then be performed (such as MS) to identify the interacting protein. AP is similar to co-immunoprecipitation; instead of immobilized antibodies, the bait protein is immobilized on the bead ( Havis et al, 2017 ). Tandem AP has a few additional steps compared to AP.…”

“…These methods are often complex, expensive, time consuming and require a significant level of expertise for performing them in a reliable manner. AP-MS and CoAP-MS (co-affinity purification mass spectroscopy) are actively studied today from separations point of view as well [39][40][41], while MS-based mapping of protein-protein interactions is progressing more slowly [42,43].…”
