Solanum tuberosum lectin inhibits Ehrlich ascites carcinoma cells growth by inducing apoptosis and G2/M cell cycle arrest

Abstract: Recently, a lectin was purified from the potato cultivated in Bangladesh locally known as Sheel. In the present study cytotoxicity of the lectin against Ehrlich ascites carcinoma (EAC) cells was studied by MTT assay in vitro in RPMI-1640 medium and 8.0-36.0 % cell growth inhibition was observed at the range of 2.5-160 μg/ml protein concentration when incubated for 24 h. The lectin-induced apoptosis in EAC cells was confirmed by fluorescence and optical microscope. The apoptotic cell death was also confirmed by… Show more

“…In vitro, exposure of EAC cells to various concentrations of MEHSR for 24 h triggered significant cell death in a dose dependent fashion ( Fig. 2a) and the results obtained from previous studies sustain the findings of this study [21], indicating the potentials of this plant extract as an anticancer agent.…”

“…Moreover, EAC cells treated with MEHSR showed characteristics features of apoptosis (cell membrane blebbing, nuclear condensation and fragmentation) whereas regular and round shape was found in EAC cells of untreated control. So characteristics of apoptotic death were observed in MEHSR treated EAC cells and similar type of observations were also reported by using different plant extracts [21,31]. All these are considered as imperative and promising characteristics of an antineoplastic agent [32].…”

“…Based on the literature, it is possible that the cytotoxicity observed in our experiments is due to apoptosis. Lectins isolated from different plants increase expression of Bax (pro-apoptotic molecule) and is dependent of caspase 3, 8, and 9 (Kabir et al, 2013, 2016; Kumar et al, 2014). It is interesting to note that lectins induce ROS production and these species is important to trigger the apoptosis mechanisms (da Mota et al, 2012).…”

“…Further analysis of mitochondrial pathway of apoptosis revealed that VC-III treatment decreased the expression of prosurvival proteins Bcl-2 and moderately increased the expression levels of proapoptotic Bcl-2 family protein, Bax. 36 Thus, VC-III induced release of cyt c into the cytosol. Studies have shown that caspase activation acts as the downstream of cyt c release during mitochondrial pathway of apoptosis.…”
