Smart Hydrogel Microfluidics for Single‐Cell Multiplexed Secretomic Analysis with High Sensitivity

Abstract: Secreted proteins determine a range of cellular functionalities correlated with human health and disease progression. Because of cell heterogeneity, it is essential to measure low abundant protein secretions from individual cells to determine single‐cell activities. In this study, an integrated platform consisting of smart hydrogel immunosensors for the sensitive detection of single‐cell secretions is developed. A single cell and smart hydrogel microparticles are encapsulated within a droplet. After incubation… Show more

“…This observation suggests that Q c had no significant impact on the diameter of droplets that were produced in the step. However, the value of Q c could influence the diameter of droplets, which were generated in the flow-focusing nozzle, and by increasing Q c , the diameter of generated droplets decreased as it could be expected 27 , 29 , 30 .…”

“…It has a potential to quantify and characterize the enzymes that secreted from single cell but it also required creating small-size droplets for single cell-based absolute quantification of enzyme [79]. The use of hydrogel immunosensors is another approach for single-cell multiplexed secretion analysis that has been proven recently [80]. The main approach to study enzymes is probably directed evolution assays.…”

“…For example, Revzin and co‐workers detected T‐cell secreted cytokines in a glass slide which were coated by patterned a polyethylene glycol (PEG) layer. Moreover, hydrogel can also be used for co‐encapsulation with single cells by emulsification (Figure D) . It works as particle immunosensors in the culture medium droplets to capture secreted cytokines for high‐resolution secretion detection.…”
