Smad7 attenuates TGF-β-mediated aging-related hypofunction of submandibular glands

Abstract: Submandibular glands have essential functions in taste, mastication, swallowing, and digestion. Submandibular gland hypofunction is prevalent in the elderly, impairing the patients’ quality of life. Current clinical treatment strategies have not decelerated or reversed the pathological process of submandibular gland hypofunction. Therefore, novel restoration strategies should be explored. However, studies on the mechanism of aging-related submandibular gland hypofunction remain very limited. The role of the TG… Show more

“…Kaplan–Meier analysis revealed a worse prognosis in the group with low KLK12 expression (Figure S6a). To validate these findings, in vitro experiments were performed by transfecting OSCC cells with small‐interfering RNAs targeting KLK12 (Hu, Liu, et al, 2021; Xie et al, 2022). The qRT‐PCR results showed a significant decrease in KLK12 expression in the si‐KLK12 group (Figure S6b).…”

“…This technique is employed to quantify the levels of specific RNA molecules within the cDNA samples. The primers used in the qPCR experiments were previously described or designed using the NCBI Primer‐BLAST design tool [ 20 ]. Subsequently, these primers were synthesized by TsingKe Biological Technology in Beijing, China.…”

“…Mouse SG samples were fixed in formalin, paraffin-embedded, and stained with Masson's trichrome, as previously described. 10 The percentage of positive staining in Masson was measured using ImageJ software ( https://cnij.imjoy.io/ ). Immunohistochemistry was performed on the formalin-fixed tissue sections using a microwave-based antigen retrieval method.…”
