Sleep Deprivation Aggravates Cognitive Impairment by the Alteration of Hippocampal Neuronal Activity and the Density of Dendritic Spine in Isoflurane-Exposed Mice

Abstract: Isoflurane contributes to cognitive deficits when used as a general anesthetic, and so does sleep deprivation (SD). Patients usually suffer from insomnia before an operation due to anxiety, fear, and other factors. It remains unclear whether preoperative SD exacerbates cognitive impairment induced by isoflurane. In this study, we observed the effects of pretreated 24-h SD in adult isoflurane-exposed mice on the cognitive behaviors, the Ca2+ signals of dorsal hippocampal CA1 (dCA1) neurons in vivo with fiber ph… Show more

“…Additionally, the impact of SD on ACC neurons in layer II/III resembles the observations in subcortical neurons, such as rat hippocampal CA1 neurons, where SD induces considerable downregulation of excitability after 72 h of SD or 24 h of sleep fragmentation [ 23 - 34 ]. However, an increase in intrinsic excitability was observed in dorsal hippocampal CA1 neurons due to 24 h SD, alongside an increase in apical dendritic spines [ 35 ]. In layer V/VI pyramidal cells of the medial prefrontal cortex, intrinsic membrane excitability increased after 8 h of SD [ 36 ].…”

“…Sleep deprivation as cognitive challenge may therefore provide a promising preclinical model of memory impairment and a useful tool to study cognition enhancing drugs ( 26 ). Previous studies exhibited that sleep deprivation could induce serious cognitive dysfunction ( 27 , 28 ). In our experiment, we found SD 2 weeks in mice could induced obviously cognitive associated behavioral impairments, such as spatial and non-spatial learning and memory deficits.…”

“…Dendritic spines are post-synaptic structures at a majority of excitatory synapses in the mammalian brain. The number and size of dendritic spines are closely related to cognitive function in different neurological diseases ( 42 ). Iso exposure was reported to lead to neuroplastic deficit.…”
