Size‐Dependant Cellular Uptake of Dendritic Polyglycerol

Abstract: To study the mechanism of cellular internalization, hyperbranched polyether derivatives consisting of amino-bearing hyperbranched polyglycerols (HPGs) of varied molecular mass and size range are designed and synthesized. HPGs were further fluorescently labelled by conjugating maleimido indocarbocyanine dye (ICC-mal). The conjugates are characterized by UV-vis spectroscopy, fluorescence profile, zeta potential, and dynamic light scattering. The uptake mechanism is studied by fluorescence-activated cell sorting … Show more

“…Amorphous atactic PDLLA cores do not promote crystallization, which resulted in spherical micelles [72]. Differences in particle topology are well known to influence cellular internalization [73,74]. Further cryo-TEM studies in combination with light scattering experiments will address these pending questions.…”

“…Of these characteristics, the particle size is a crucial factor which determines NP endocytosis pathway [17,18], uptake rate and efficiency [6,16,[19][20][21][22], in vitro cytotoxicity [23][24][25][26][27][28], immune responses [27][28][29][30], and the final localisation [15,31,32]. Thus size-dependent uptake of various NPs has been intensively investigated and widely reported.…”

“…The carrier was rationally designed and synthesized to bear a MW of 200 kDa, previously reported to be within the size range exploited better accumulation and cellular uptake (Reichert et al, 2011). The FRET quenching efficacy was 94% of the intact theranostic probe, and terminated upon cell internalization and the subsequent cleavage, mostly inside the low-pH acidic, endolysosomal system.…”
