Site-specific integration of the temperate bacteriophage phi adh into the Lactobacillus gasseri chromosome and molecular characterization of the phage (attP) and bacterial (attB) attachment sites

Abstract: The temperate bacteriophage +adh integrates its genome into the chromosomal DNA ofLactobaciUus gasseri ADH by a site-specific recombination process. Southern hybridization analysis of BclI-digested genomic DNA from six relysogenized derivatives of the prophage-cured strain NCK102 displayed phage-chromosomal junction fragments identical to those of the lysogenic parent. The +adh attachment site sequence, attP, was located within a 365-bp EcoRI-HindIII fragment of phage 4adh. This fragment was cloned and sequenc… Show more

“…These data also suggest that V. cholerae strains have the potential to acquire, excise, and reintegrate the VPI and that strains in which the VPI has excised retain the attL site sequence in their genome. It is noteworthy that the mechanisms described above for the VPI are typical of lysogenic and temperate phages (41). The relevance of this specific sequence is described below.…”

“…Besides the VPI, a PAI of Salmonella enterica serovar Typhimurium is also inserted into the ssrA gene (23). Insertion of PAIs into tRNA or tRNA-like genes is typical of many pathogenic bacteria (3), and this feature is also shared among lysogenic and temperate phages (19,41,42).…”

“…Lactic acid bacteria were transformed by electroporation in 0.2 cm cuvettes in a Gene Pulser apparatus (Bio-Rad; settings 25 pF, 2.5 kV, 200 Q). Electro-competent cells were prepared according to the method of Raya et al (1992). Hybridization.…”

“…Two temperate phages from L. delbrueckii (145,146) and L. gasseri (147) were used to construct site-specific integration vectors. A DNA sequence was stably introduced into the chromosome of L. casei by application of prophage integration and it could be subsequently excised (148).…”
