Single short-term signal that enhances cooperative activity of old rat hepatocytes acts for several days

Abstract: Primary cultures of rat hepatocytes were studied in serum-free medium. Ultradian protein synthesis rhythm was used as a marker of overall cell synchronization and cooperation amongst the population. The level of synchronization was determined by amplitudes of the rhythm. Low synchronization of old rat hepatocytes can be enhanced by addition of either gangliosides or phenylephrine to the medium. Incubation of cultures with gangliosides lasted for 2.5 h, while action of phenylephrine was only for 2 min. The ampl… Show more

“…cooperation of cells in the formation of a common rhythm in a cell population. The main data have been published during the last five years (Brodsky et al, a, b, 2004(Brodsky et al, , 2005Zvezdina et al, 2000). The ultradian rhythm of protein synthesis was used as a marker of cell interactions and synchronisation of oscillations in rat hepatocytes cultured on slides.…”

“…Ultradian rhythms are an innate cell property presumably of a fractal nature (Barnsley et al, 1987;Brodsky, 1992Brodsky, , 2006Lloyd, 1992Lloyd, , 2005Lloyd, 1993, 1995). In a series of articles already published in this journal (Brodsky et al, 2000(Brodsky et al, , 2003a(Brodsky et al, ,b, 2004(Brodsky et al, , 2005, some signal factors that promoted synchronization of protein synthesis rhythm were described. Depending on cell density, two types of primary hepatocyte cultures on slides in the serum-free medium were obtained, dense cultures with closely arranged cells, and sparse cultures with widely separated cells.…”

“…It was clear that experimental synchronization of sparse cultures as well as desynchronization of dense cultures could be considered as actual events for the analysis of cell self-organization. In a series of articles already published in this journal (Brodsky et al, 2000(Brodsky et al, , 2003a(Brodsky et al, ,b, 2004(Brodsky et al, , 2005, some signal factors that promoted synchronization of protein synthesis rhythm were described. The cells responded to either ganglioside or catecholamine signaling with an increase of calcium ions in the cytoplasm.…”
