Single platelet variability governs population sensitivity and initiates intrinsic heterotypic responses

Abstract: Investigations into the nature of platelet functional variety and consequences for homeostasis require new methods for resolving single platelet phenotypes. Here we combine droplet microfluidics with flow cytometry for high throughput single platelet function analysis. A large-scale sensitivity continuum was shown to be a general feature of human platelets from individual donors, with hypersensitive platelets coordinating significant sensitivity gains in bulk platelet populations and shown to direct aggregatio… Show more

“…Reintroducing paracrine signaling between platelets increased platelet population reactivity 20-fold. 72,73 This implies the presence of hyper-reactive platelets with a pioneer hemostatic function, although this remains speculation without experiments linking such platelets to thrombus initiation. With continued thrombus growth, the phosphatidylserine-exposing platelet polarization phenomena occurs, thus producing proaggregatory and procoagulant states.…”

“…Furthermore, Jongen et al (2020) combined droplet microfluidics with flow cytometry for high-throughput single platelet function analysis ( Figure 1H ). The device incorporated four individual inlets to infuse platelets, agonist/antagonist solution and fluoro-oil, which eventually encountered at a common junction where analytes were encapsulated within the fluoro-oil droplets.…”

“…Platelets are small yet highly abundant blood cells that contribute to hemostasis, immunity, and inflammation [1]. Once considered homogeneous, they are now recognized as heterogeneous in size, age, receptor expression, and response to agonist stimulation [2]. While the molecular origins of platelet heterogeneity are unclear, they are likely driven by developmental and environmental factors.…”
