Simultaneous quantification of twelve active components in Yiqing granule by ultra‐performance liquid chromatography: application to quality control study

Abstract: An ultra-performance liquid chromatography (UPLC) method coupled with photodiode array (PDA) detection has been developed and validated for simultaneous quantification of 12 active components (berberine, palmatine, jatrorrhizine, aloe-emodin, rhein, emodin, chrysophanol, physcion, baicalin, baicalein, wogonoside and wogonin) in Yiqing granule. Optimum separation were achieved on a C(18) column (50 × 2.1 mm i.d., 1.7 µm particle) through a 7.5 min gradient delivery of a mixture of A (acetonitrile) and B (0.1% a… Show more

“…Many studies have mainly focused on the analysis of sesquiterpenoids and phenylpropanoids from ATS (Lao et al ., ; Venskutonis and Dagilyte, ; Yu et al ., ; Lee et al ., and ; Deng et al ., ). Since high‐performance liquid chromatography–mass spectrometry (HPLC‐MS) has become a powerful tool for qualitative analysis of TCM and metabolites owing to its high resolution, accuracy and reproducibility (Ling et al ., ; Fu et al ., ; Chen et al ., ; Bagnall et al ., ; Martinez Bueno et al ., ; Qi et al ., ), we have developed a method using a high‐performance liquid chromatography coupled with electrospray ionization quadrupole time‐of‐flight mass spectrometry (HPLC/ESI‐QTOF‐MS/MS) technique for a comprehensive qualitative analysis of ATS. In addition, few studies in quantifying other types of constituents of ATS have been reported to date.…”

“…Yiqing Capsule is a medicinal preparation developed on the basis of the well-known TCM formula named San Huang Xie Xin decoction. This herbal preparation is composed of three Chinese medicinal herbs including Rhizoma Coptidis (Coptis chinensis), Radix et Rhizoma Rhei (Rheum officinale) and Radix Scutellariae (Scutellaria baicalensis), and is commonly used in clinical practice for the treatment of intense heat in the body, inflammation and painful swelling of the eyes, sore throat, gingival bleeding, reddish urine, constipation, pharyngitis and amygdalitis [1]. There are over 40 manufacturers that produce commercial Yiqing preparations in either granule or capsule across China.…”

“…By now, some analytical methods have been developed for the quantitative analysis of the target components in Yiqing Capsule to assess the quality of the complex preparations [1][2][3][4][5]. However, low sensitivity and possible uncertainty of chromatographic peak identification (mainly by retention time) made high-performance liquid chromatography (HPLC)-UV methods unlikely match the routine analysis of interested compounds, especially for the minor ones in complex plant extracts.…”

“…And this strategy has been successfully applied for assessing the quality of TCM preparations, Qin-Bao-Hong antitussive tablet, Yiqing granule and Wuji pill. [10][11][12] In the implementation of this strategy, it is difficult for all the markers to obtain good separation in a single run. And the degree of this difficulty would increase with the varieties of constituent medicinal herbs.…”

“…13,14 In the previous study, UPLC was employed for TCM preparations in which the variety of compositions was less (three constituent medicinal herbs), and showed excellent separation efficiency, large peak capacity, high resolution, short retention time, low solvent consumption and high sensitivity. 11,12 Furthermore, we try to employ this strategy and UPLC to the quality control of TCM preparations with more varieties of constituent medicinal herbs. Wuhu powder is a TCM preparation comprising ve Chinese medicinal herbs, i.e., Radix Angelicae Sinensis, Flos Carthami, Radix Saposhnikoviae, Rhizoma Arisaematis Preparatum and Radix Angelicae Dahuricae.…”
