A simple, sensitive and rapid spectrophotometric method was developed and validated for the determination of two skeletal muscle relaxants namely, tizanidine hydrochloride (I) and orphenadrine citrate (II) in pharmaceutical formulations. The proposed method is based on the formation of a binary complex between the studied drugs and eosin Y in aqueous buffered medium (pH 3.5). Under the optimum conditions, the binary complex showed absorption maxima at 545 nm for tizanidine and 542 nm for orphenadrine. The calibration plots were rectilinear over concentration range of 0.5-8 μg/mL and 1-12 μg/mL with limits of detection of 0.1 μg/mL and 0.3 μg/mL for tizanidine and orphenadrine respectively. The different experimental parameters affecting the development and stability of the complex were studied and optimized. The method was successfully applied for determination of the studied drugs in their dosage forms; and to the content uniformity test of tizanidine in tablets.

A sensitive spectrofluorimetric method was developed for the determination of tizanidine in human plasma, urine and pharmaceutical preparations. The method is based on reaction of tizanidine with 1-dimethylaminonaphthalene-5-sulphonyl chloride (dansyl chloride) in an alkaline medium to form a highly fluorescent derivative that was measured at 511 nm after excitation at 383 nm. The different experimental parameters affecting the fluorescence intensity of tizanidine was carefully studied and optimized. The fluorescence-concentration plots were rectilinear over the ranges 50-500 and 20-300 ng/mL for plasma and urine, respectively, detection limits of 1.81 and 0.54 ng/mL and quantification limits of 5.43 and 1.62 ng/mL for plasma and urine, respectively. The method presents good performance in terms of linearity, detection and quantification limits, precision, accuracy and specificity. The proposed method was successfully applied for the determination of tizanidine in pharmaceutical preparations. The results obtained were compared with a reference method, using t- and F-tests.

Synthesis and characterization of core–shell magnetic molecularly imprinted polymer nanoparticles for selective extraction of tizanidine in human plasma

In this study, simple, effective and general processes were used for the synthesis of a new nanomolecularly imprinted polymers (MIPs) layer on magnetic Fe 3 O 4 nanoparticles (NPs) with uniform core-shell structure by combining surface imprinting and nanotechniques. The first step for the synthesis of magnetic NPs was co-precipitation of Fe 2+ and Fe 3+ in an ammonia solution. Then, an SiO 2 shell was coated on the magnetic core with the Stöber method. Subsequently, the C=C groups were grafted onto the silica-modified Fe 3 O 4 surface by 3-(trimethoxysilyl) propyl methacrylate. Finally, MIPs films were formed on the surface of Fe 3 O 4 @SiO 2 by the copolymerization of C=C end groups with methacrylic acid (functional monomer), ethylene glycol dimethacrylate (crosslinker), 2,2-azobisisobutyronitrile (initiator) and tizanidine (template molecule). The products were characterized using techniques that included Fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), thermo gravimetric analysis (TGA), scanning electron microscopy (SEM), UV spectrophotometry, transmission electron microscopy (TEM) and vibrating sample magnetometer (VSM). Measurement of tizanidine through use of the coreshell magnetic molecularly imprinted polymers nanoparticles (MMIPs-NPs) in human plasma samples compared to the paracetamol showed that the synthesized nanosized MMIP for tizanidine has acted selectively.
