Chlorpheniramine maleate–paracetamol–caffeine tablet formulation is one of the common over-the-counter drugs used for the treatment of cold and cough. A reversed-phase high-performance liquid-chromatography method has been successfully developed for the simultaneous determination of chlorpheniramine maleate, paracetamol and caffeine in a drug formulation. The RP-HPLC method employed a Phenomenex C18 reversed phase column (Luna 5µ, 250 × 4.6 mm) with an isocratic mixture of methanol and 0.05 M dibasic phosphate buffer pH 4.0 in the ratio of (30:70; v/v) as the mobile phase. The column temperature was kept at 30 °C. The flow rate was 1.0 mL/min and detection was by means of a UV detector at wavelength of 215 nm. All the active components were successfully eluted with mean retention times of 2.4, 4.2, 7.2 min for chlorpheniramine maleate, paracetamol and caffeine respectively. The method was found to be linear (R2 > 0.99), precise (RSD < 2.0 %), accurate (recoveries 97.9–102.8 %), specific, simple, sensitive, rapid and robust. The validated method can be used in routine quality control analysis of fixed dose combination tablets containing chlorpheniramine maleate, paracetamol and caffeine without any interference by excipients.

Derivative spectrophotometric (first, second, third and fourth derivative) were developed for the determination of Sulphamethoxazole)SMX) and trimethoprim (TMP)antibiotic binary mixture by applying zero-crossing technique for pure synthetic mixture while simultaneous determination of (SMX) and (TMP) in tabletsusing D 4 at waveleanth 257.8 nm and 251.5 nm for (SMX) and (TMP) the correlation coefficient of calibration curves were 0.9992 and 0.9995. Linearity is maintained within a wide concentration range from 2.00 to 25.00 mg/l, with relative error (-0.11 and 0.60) the LOD (0.360 and 0.382 mg/L) and LOQ and (1.200 and1.275 mg/L) respectively.No interference found between both determined and those of matrices. A good accuracy and precision of simultaneous determination of (SMX), and (TMP) were confirmed by statistical analysis.The percentage recovery of the individual drugs under the established conditions is ranged from 97.23% to 102.13 %. This method was compared with the British pharmacopoeiamethodusing F test. The procedures do not require any separation step.

The objective of this study is to evaluate the capability of UV-spectrophotometry in combination of multivariate calibration based on Partial Least Square (PLS) regression for simultaneous quantitative analysis and dissolution evaluation of sulfamethoxazole (SUL) and trimethoprim (TRI) in tablet dosage form. The experimental calibration and validation matrixes were constructed with 20 and 10 samples, respectively. The concentration range considered was 1-16 µg mLG 1 for both SUL and TRI. The absorbance data of the calibration standards were taken between 200-400 nm. For achieving the best calibration model, the related parameters were evaluated. The optimum number of factors was selected by using the cross-validation method. The evaluation of calibration model is relied on the coefficient of determination (R 2 ) and Root Mean Square Error of Calibration (RMSEC). The coefficient of determination (R 2 ) for the relationship between actual values and predicted values of SUL and TRI was higher than 0.99 indicating good accuracy of the developed method. The RMSEC values obtained were relatively low, namely 0.167% (SUL) and 0.279% (TRI), which indicate acceptable precision of analytical method. The accuracy of developed method was comparable to that of High Performance Liquid Chromatography (HPLC) method. The UV spectrophotometry in combination with PLS calibration model was successfully used for quantitative analysis and dissolution studies of SUL and TRI sulfamethoxazole and trimethoprim in tablet dosage form.
