Simultaneous Determination of Strychnine, Brucine, Strychnine N-Oxide and Brucine N-Oxide in Crude and Fermented Nux Vomica by CZE with Ephedrine Hydrochloride as an Internal Standard

Abstract: A rapid capillary zone electrophoresis (CZE) method with ephedrine hydrochloride as an internal standard was developed and validated for separating and quantifying the alkaloids strychnine (1), brucine (2), strychnine N-oxide (3), and brucine N-oxide (4) in the seed of Strychnos nux-vomica L. (crude nux vomica) and its fermented products by fungus Trametes cinnabarina (Jacq.: Fr.) Fr. (fermented nux vomica). The contents of alkaloids (1 and 2) in crude nux vomica were higher than those in fermented drugs, wher… Show more

“…This analytical method with low intra‐ and interday variations had detection limits of 0.83 ppm for strychnine, 0.96 ppm for brucine, 2.10 ppm for strychnine N ‐oxide, and 2.50 ppm for brucine N ‐oxide. Strychnine N ‐oxide and brucine N ‐oxide were found only in fermented nux vomica, which indicates that strychnine and brucine in crude nux vomica might be partly transferred to their N ‐oxides .…”

“…In the next work, CE–ELCL was applied to monitor the concentration of toxic diester‐diterpenoid alkaloids in Radix Aconiti . Lastly, CZE–UV was employed for the simultaneous determination of strychnine, brucine, strychnine N ‐oxide, and brucine N ‐oxide in crude and fermented nux vomica . It is noted that for human consumption, crude nux vomica containing toxic strychnine and brucine must be processed by fermentation to their less toxic N ‐oxide derivatives.…”

“…Semen Strychni is a traditional Chinese herb, mainly used to treat rheumatoid arthritis, nervous disease, and traumatic pains and to promote blood circulation and remove blood stasis [ 1 – 3 ]. Total alkaloid from Semen Strychni (TAS) is a concentrated bioactive extract of Semen Strychni , which mainly consists of strychnine and brucine [ 4 , 5 ]. Strychnine and brucine possess analgesic, anti-inflammatory, and antitumor effects as well as toxicity [ 6 , 7 ].…”
