Long-term treatment with glucocorticoids induces weight gain and increased risk to develop obesity-related metabolic complications. The underlying mechanisms are not fully understood. Glucocorticoid therapy has previously been associated with increased levels of circulating leptin. In this study the eating behaviour was therefore studied in relation to leptin levels before and after shortterm prednisolone treatment. Design. Within-subject design. Subjects. Twelve healthy postmenopausal women with a mean body mass index (BMI) of 28.9 kg m )2 (±0.8 SEM) volunteered after recruitment by an advertisement in the local paper. Interventions. The subjects received 25 mg prednisolone daily for 7 days. Main outcome measurements. Fasting serum samples were obtained before, during and after treatment for determination of leptin and insulin, glucose and fractionated lipoproteins in plasma. The microstructure of the eating behaviour was registered with a universal eating monitor, VIKTOR. Appetite was estimated by visual analogue rating scales and food intake by a 48-h recall. Results. Serum leptin increased after 2 and 7 days of glucocorticoid administration (P < 0.01), and the food intake measured by VIKTOR after 7 days of treatment (P < 0.05). No statistically significant changes were however, found in the 48-h foodrecall or in the subjective appetite registrations. Insulin levels were borderline elevated (P ¼ 0.062) after treatment, but no significant changes of fasting glucose were seen. High density lipoprotein cholesterol (HDL) increased (P < 0.05), whilst low density lipoprotein cholesterol (LDL) decreased (P < 0.05). Conclusion. Food intake was elevated after glucocorticoid administration as observed with an objective, quantitative method, in spite of increased levels of circulating leptin.

The aim of this study was to verify the effect of nasally instilled liposomes (L) or dexamethasone-containing L (Ldexa) on normal or inflamed lung tissue. Three groups of mice were studied. Group I was given saline instillations for 3 weeks prior to the instillation with liposomes. In groups II and III lung inflammation was induced by repeated instillations of Saccharopolyspora rectivirgula before the instillation of liposomes (group II) or liposomes containing dexamethasone (group III). Animals from all groups were killed at regular time intervals for up to 48 h after the instillation of liposomes. The total cell count in bronchoalveolar lavage fluid did not differ significantly between groups I and II. However, in group III it decreased rapidly from 6.2 to 2.8 x 10(5) cells mL-1 within 2 h. Differential counts did not change in group I, but in group II a transient neutrophilia was observed 180 min after the instillation of liposomes. In group III, the instillation of dexamethasone-containing liposomes depleted all neutrophils and lymphocytes after 4 h. No TNF alpha was found in samples of lavage fluid from any of the groups at time 0. In group I, liposomes induced the production of 0.03 ng mL-1 of TNF alpha in the 1 h sample only. In group II, TNF alpha peaked to 1 ng mL-1 at 1 h and had decreased to 0.35 ng mL-1 by 3 h. In group III, TNF alpha peaked at 1 h, but only reached a level of 0.1 ng mL-1.(ABSTRACT TRUNCATED AT 250 WORDS)

In this paper we present introduction and development of some new analytical methods for identification of anabolic steroids, their metabolites and certain hormones, especially determination of exogenous testosterone by means of gas chromatography-mass spectrometry. Identification of central nervous stimulants and corticosteroides has been performed by high performance liquid chromatography. In desire to achieve better results, to increase strength and endurance, to sharpen reflexes and to reduce stress and anxiety athletes as well as other people use different pharmacological substances, hormones or even illicit drugs. Use of these substances without medical supervision can lead to adverse effects to one's health or even cause a death. At the same time, use of such substances means a kind of cheat that could not be accepted. This is why International Olympic Committee started at 1968 with official doping control that is permanently carried out and continuously increasing number of banned substances. Doping control demands for discover and development of new sensitive and specific methods for detection of banned substances and their metabolites in urine and blood.
