Simultaneous determination of levofloxacin and ciprofloxacin in microdialysates and plasma by high-performance liquid chromatography

a b s t r a c tHerein, we developed a novel integrated device to perform phase separation based on ultrasound-assisted salt-induced liquid-liquid microextraction for determination of five fluoroquinones (FQs) in human body fluids. The integrated device consisted of three simple HDPE components used to separate the extraction solvent from the aqueous phase prior to retrieving the extractant. A series of extraction parameters were optimized using the response surface method based on central composite design. Optimal conditions consisted of 945 L acetone extraction solvent, pH 2.1, 4.1 min stir time, 5.9 g Na 2 SO 4 , and 4.0 min centrifugation. Under optimized conditions, the limits of detection (at S/N = 3) were 0.12-0.66 g L −1 , the linear range was 0.5-500 g L −1 and recoveries were 92.6-110.9% for the five FQs extracted from plasma and urine. The proposed method has several advantages, such as easy construction from inexpensive materials, high extraction efficiency, short extraction time, and compatibility with HPLC analysis. Thus, this method shows excellent prospects for sample pretreatment and analysis of FQs in human body fluids.© 2015 Elsevier B.V. ABSTRACT:Fluoroquinolones are the most commonly prescribed synthetic antibacterial agents having improved pharmacokinetic properties and broad spectrum of activity. Due to their broad clinical activity they are the most promising drug for antibacterial chemotherapy. The current study focused on the application of similar chromatographic system for the analysis of a number of fluoroquinolones from bulk, pharmaceutical formulations and biological matrices. We have also summarized how a single chromatographic system can be used for the quantification of a number of fluoroquinoloes from their respective marketed formulation. A detailed systemic survey of the sample preparation techniques, the chromatographic conditions and detection of the respective fluoroquinolones has been presented in this study. In the course of this discussion the advantages and applicability of the high performance liquid chromatographic techniques are also examined.

Resonance Rayleigh scattering approach based on association complex formation with erythrosine B for determination of venlafaxine, application to the dosage form and spiked human plasma

The interaction of venlafaxine hydrochloride (VLX) with erythrosine B was investigated using a resonance Rayleigh scattering (RRS) spectroscopic technique. In acetate buffer (pH 3.4), erythrosine B reacted with VLX to form a 1:1 ion-pair complex with concomitant enhancement in RRS intensity that was measured at 330 nm. In addition, the stability constant and the change in free energy of the reaction were estimated. Based on this interaction a new method was developed for a sensitive VLX analysis using erythrosine B as a probe. The results indicated that this method had good selectivity in the presence of coexisting compounds. The scattering intensity (ΔI RRS ) was linearly dependent on VLX concentration over the range 0.04-1.0 μg ml À1 with a determination coefficient (r) of 0.9998. The limit of detection and limit of quantitation were 0.01 and 0.03 μg ml À1 , respectively. This method could be suitably used for analysis of VLX in pharmaceutical capsules and human plasma.
